چکیده

Background. Prolonged membrane depolarization induced by an electric shock in the heart may produce propagation block leading to repetitive beats. We studied prolonged depolarization and its role in repetitive beats in a thin epicardial layer of endocardially prefrozen arterially perfused rabbit heart. Methods and Results. A laser scanner recorded optical action potentials at 63 sites within a 1-cm2 area on the left ventricle of hearts stained with potentiometric fluorescent dye. Pacing (S1) produced propagation across the myofibers; then, a 3-millisecond shock (S2) given in the S1 refractory period produced an electric field that decreased in strength with distance along the fibers. The S2 strengths at the center of the scanned region (C) were 2.1±0.2 or 5.6±03 V/cm (mean±SD, n=4). Repetitive beats occurred in 50% of hearts when C was 2.1 V/cm and in 100% of hearts when C was 5.6 V/cm. With each occurrence of repetitive beats, prolonged depolarization of the shocked action potential occurred within 1

Proarrhythmic effects of flecainide. Experimental evidence for increased susceptibility to reentrant arrhythmias.

BACKGROUND The goal of this study was to investigate the nature and electrophysiological mechanisms of the proarrhythmic effects of flecainide in Langendorff-perfused rabbit hearts. METHODS AND RESULTS A thin layer of epicardium was obtained by an endocardial cryotechnique in 10 Langendorff-perfused rabbit hearts. Six other hearts were kept intact. Programmed electrical stimulation using up t...

Dispersion-based reentry: mechanism of initiation of ventricular tachycardia in isolated rabbit hearts.

The aim of the study was to determine whether facilitation of reentry by potassium-channel openers is related to dispersion of refractoriness and/or modification of anisotropic properties of ventricular myocardium. The dispersion of ventricular effective refractory period (VERP), longitudinal and transverse ventricular conduction velocities (θL and θT, respectively), and wavelength [λ = VERP × ...

Polymorphic reentrant ventricular tachycardia in the isolated rabbit heart studied by high-density mapping.

BACKGROUND The role of dispersion of refractoriness and reentry for the genesis of polymorphic ventricular tachycardia (VT) has recently become emphasized. We investigated the mechanisms of polymorphic arrhythmias in a 2D preparation confining an area of prolonged refractoriness. METHODS AND RESULTS In 16 Langendorff-perfused rabbit hearts, a sheet of left ventricular epicardium was obtained ...

The role of Na+-K+-ATPase in the basic and rate-dependent properties of isolated perfused rabbit Atrioventricular Node

Introduction: Ouabaine is a well-known atrioventricular (AV) node depressant agent, but its effects on functional properties of the AV node have not been cleared. The aim of the present study was to determine how ouabaine administration modifies the rate-dependent properties of the AV node. Methods: Selective stimulation protocols were used to quantify independently electrophysiological prop...

The Role of Anion Exchanger on Pulmonary Vascular Response to Sustained Alveolar Hypoxia in the Isolated Perfused Rabbit Lung

Background: Some respiratory diseases may induce alveolar hypoxia thereby hypoxic pulmonary vasoconstriction (HPV). However, the mechanisms of this physiologic phenomenon are not fully understood. This study was the first to investigate the role of anion exchanger in sustained HPV.Methods: Experiments were performed in the isolated perfused rabbit lung. After preparation, the lungs were divided...
